This procedure has been established for checking the effectiveness of UV-light used in laminar airflow cabinet and pass through window of sterile manufacturing and testing area
Scope:
This procedure is applicable to sterile manufacturing area and microbiology lab.

Microbiologist.

Quality Control Manager.
Procedure:
Prepare nutrient agar as per SOP for preparation of culture media.
Aseptically pour approximately 15–20 ml of nutrient agar previously sterilized transfer 1ml containing 105 cfu per ml culture of Bacillus subtilis to two nutrient agar plates & swirl the plate gently to mix properly and allow solidifying.
After solidifying switch on UV-light for 30 minutes and open the lid and set the plate under UV-light under laminar airflow cabinet.
After 30 minutes exposure, switch "OFF" the UV-light & close the lid of nutrient agar plate.
Incubate both the nutrient agar plates in the hot incubator at 32oC±2.5oC for
24-48 hours.
Place one agar plate un-expose as positive control in the incubation and negative control without culture and repeat the same procedure for UV-light of pass through box.
At the end or completion of incubation period observe the plate for any sign of total bacterial growth. Record the observations on respective form.
Frequency: Once in the month
Acceptance Criteria:
There must not be any sign of growth in the plate exposed under UV-light and negative control and growth must be observed in the positive control
