Objective:
This procedure has been established for for swab sample collection from the equipment's and surfaces of floor and walls.
Scope:
This procedure is applicable to all the areas where product is being manufactured and tested i.e. (production areas and testing areas)

Procedure:
Sampling Method For Microbiological Analysis
Take a pre-sterilized swab stick and transfer under laminar airflow cabinet and pour aseptically about 5-10ml of sterilized distilled water or WFI .
After preparing the swab stick for sampling mark it properly with the serial number and name of location and wrap it in aluminum foil and move to the sampling area.
Then unpack the swab stick and remove the swab and in unidirectional move
(Not to and fro motion) swab the surface of equipment or floor in an area of
10 x 10 sq./cms.
After sampling close the swab and wrap it in aluminum foil and move to microbiology lab. Under laminar airflow cabinet transfer the solution in 100ml of the buffered peptone water previously sterilized. set the media for 15 minutes. and then perform microbial limit test as per SOP
Sampling Method For Chemical Analysis
Take a swab stick and pour 5ml of distilled water.
After preparing the swab stick for sampling mark it properly with the serial number and name of location and move to the sampling area.
Then take the swab of the surface of equipment or floor in an area of 10 x 10 sq./cms. and move the swab to QC department.
In QC lab pour the swab sample solution into the 10ml of Acetate Buffer pH 4 in 100ml of volumetric flask and sonicate the solution for 15 minutes.
After sonication make up the final volume upto 100ml with acetate buffer pH 4
Analyze this solution as per the specifications
